PART III: HYDROPHILIC INTERACTION LIQUID CHROMATOGRAPHY (HILIC)
Hydrophilic interaction liquid chromatography (HILIC) is a modern and dynamic separation method. This tech-
nique enables the study of highly polar substances, impossible to analyze the normal- and reversed-phase systems,
due to the irreversible adsorption or absence of interactions with the stationary phase, respectively. HILIC system
consists of a highly hydrophilic stationary phase, ie. unmodified silica or chemically modified silica that includes
cyjano, hydroxyl or amine groups. The mobile phase is a mixture of a polar organic solvent (acetonitrile) with
water content up to 40%. The analysis conditions in the HILIC system determine the presence of the specific
retention mechanism. The hydrophilic interaction liquid chromatography retention of analytes base on the distri-
bution of analyte between the mobile phase and water layer adsorbed on the surface of the stationary phase. In
recent years HILIC system is widely used in various research and chromatographic analyzes.
